Personality:
First off, he's a doctor so he's very dedicated to saving all life in the universe. He may not particularly care for certain races, such as Klingons or Romulans, but he will treat them to the best of his abilities. Unfortunately, his bedside manner is lacking compassion and he rarely lets his patients do anything without his say-so. That includes sitting upright. Doctor knows best, after all. He's very dedicated to his craft. In the book, Crisis on Centaurus, we got a bit of insight on that and learned how him met Jim. He passed up several Shore Leaves from his assignment to treat and rehabilitate the, then, Ensign Kirk after he shattered his knee and part of his leg. Shore leaves that would have been spent with his nine year old daughter. Now that is dedication!
In general though, he comes off as a very gruff old man with an intense phobie of transporters. If possible, he will always take a shuttle down to any planet or to a space station. Sadly, he rare has that opportunity. To his death, he will always remain suspicious of technology that de-atomizes a person. He said it best himself in the episodes 'Space Seed': "I signed on this ship to practice medicine, not to have my atoms scattered back and forth across space by this gadget!"
Bones can be very courageous. That isn't to mistake he isn't fearful, any man would be afraid of certain situations, such as when Khan held a knife to his throat but Bones remained as cynical and sacarstic as he tends to be. And he'll save any man that's hurt, even enemies (as I mentioned earlier). His loyalty to his friends and shipmates is one of his most important qualities. Spock may not think so often, but Bones does put a lot of thought into the things he does and potential outcomes. His temper is famous, certain things set him off in a heartbeat and it takes either the coldly logical Spock to talk reason to him or his captain, and closest friend, Jim.
He's a blunt man who rarely sugarcoats anything he has to say. Even when it's a superior officer because, by God, he will not let something happen if he could convince or scold someone about any absolutely stupid decisions they might make. Kirk happens to be a common target.
Leonard McCoy is a man of many talents, he can mix drinks, outdrink any man (except maybe Scotty or Spock), has the "hands of a surgeon" according to the Dax symbiont, never peeks unless it's in the line of duty and acts as the ships psychologist. He's also quite intelligent, necessary for anyone who goes through Starfleet Academy and is assigned to starships, and was one of the men responsible for curing the common cold.
He takes particular entertainment from ribbing the ships First Officer, often remarking on him being green-blooded or being a cold logical Vulcan. He takes whatever insults he can get but don't let that fool anyone. He has a deep respect for the Vulcan officer and even calls him friend. Kirk and Spock are two men he would give his life to save, if ever they were injured fatally.
First Person:
God--
[Eyes the watch on the ground and scoops it up. Looking it over until he triggers the video function. Now... Allow him to be an old grump at you, city-dwellers.]
What in th'hell is this? Now I know I wasn't headin' for no away missions so I sure as hell know Scotty didn't beam onto that damn relic! I expect a damn good explanation for this!
[Picking up his Tricorder, a large boxy-type of Medical Tricorder specifically, and the grabs a small medkit he'd been brought with.]
I'm a damn doctor, not a guinea pig for some ridiculous transporter experiment! They damn well know I don't trust them things.
[Grump grump grump.]
Third Person:
"You know, I ain't one for surprises, and this is a surprise I could live without. Joanna visitin' the Enterprise, I can live with. Scotty shuttin' down the engines for an overhaul, fine as long as I got power in Sickbay. Endin' up on some godforsaken world with no contact to Starfleet? I'll be damned if I sit by and take this nonsense!" He's sitting in the clinic, looking over the equipment and looking very disappointed by it all.
"If you're gonna bring me here, you could at least provide me with medical tools that aren't from the damn Dark Ages!" In comparison to his time, anyway. "My medkit is not going to cut it with what I heard this city goes through! I expect a little more help here than what Nurse Chapel left behind! I don't give a damn how powerful you think you are, if you can bring a single man here from a starship, you can damn well bring some of my equipment!"
Rant rant, rage, grr!
"God, I need a drink..." Off to the bar!

He never peeks unless it's "in the line of duty" and acts as the ships psychologist. He's also quite intelligent, necessary for anyone who goes through Starfleet Academy and is assigned to starships, and was one of the men responsible for curing the common cold, later on in life.

Kirk happens to be a common target. He's very snarky, always giving Jim or Spock a hard time but it's all in good faith that they understand where he's coming from and he's rarely reprimanded for it. Apparently Starfleet appreciates someone who can be painfully honest. Basically, he's sharp-tongued, skeptical, foul-mouthed, and he won't hesitate to speak his mind, no matter how unpopular or controversial of an opinion he might be voicing. And he has a right to be like that. He was married and his marriage fell apart because his wife didn't approve of his career choices, and in that divorce, he pretty much lost his daughter. He got custody, yes, but he refused to raise her on a Starship, so he sent her to live with his sister. It was one of the most painful decisions in his life.
An optimist, even after his divorce, Bones continued his work and created a few friendships that would last until he peacefully dies, only a few weeks after inspecting the Enterprise D. However part of that optimism comes with a dark side. His hopes are always up, looking for the best conclusion so his world shatters bit by bit when something happens. His foul attitude is testament to that.

In general, Bones is a very emotional man. He gets pissed at Spock for doing something stupid and he gets pissed at Spock for one-upping him. He enjoys challenging Spock to a game of wits and definitely enjoys watching one of those few occasions that Jim beats Spock in chess or 3D chess. Jim is his closest friend and there were times that he would have given up his life for his friend and captain. He was faithful to Spock when he implanted his Katra (while he's not at that point in canon yet, it shows just how faithful he can be) into his mind and recovered his resurrected body and both went to Vulcan to put Spocks' mind where it belongs. In a way, Bones is the direct opposite of Spock: He is emotional and persistent while Spock rejects his emotions and pursues something only if logic dictates it.
He plays up the role of Southern gentleman to the point of sometimes appearing too laid back to be serious. Friendly as he can be though, his only close friends are Jim Spock. His relationship with his daughter is woefully lacking, due to his career and how few times he was near enough to Centaurus to visit her. He hasn't spoken with his wife in years, since their divorce, so he obviously hasn't got a good relationship with her either.

"God, I need a drink..." He didn't want a drink. He legitimately needed one. So, off to the bar he went! Hopefully he could find something a little stronger than scotch. He wanted something that would put hairs on his chest and knock him on his ass. He'd mix one himself, if he had to.
